چکیده

Transient receptor potential vanilloid 4 (TRPV4) is a Ca2+-permeable non-selective cation channel in TRP family and highly expressed skin tissue. Several studies have demonstrated its multiple function the regulation of inflammation, fibrosis tumor growth. We recently identified that TRPV4 expression was increased wounded area. However, role cutaneous wound healing not well characterized, therefore we aimed to investigate it. Cutaneous experiment performed WT KO mice. Skin tissues from area were collected for histological biochemical studies. Mouse primary keratinocytes derived mice NIH3T3 cells usedin vitro study. Wound significantly delayed 2 days after wounding (p<0.01) compared with Pathological revealed impaired re-epithelialization formation granulation tissue Protein mRNA levels type I collagen αSMA decreased (p<0.05). In vitrostudy, migrative capacity mouse (p<0.01). Immunoblotting assay scratch stimulus enhanced ERK phosphorylation level suppressed by antagonist (HC-067047) administration. Elevated COL1A1 ACTA2 TGF-β stimulated treatment. conclusion, promotes migration via activation MAPK signaling pathways induced myofibroblasts differentiation, resulting accelerated healing. The channels may therapeutic intractable wounds.

Vimentin coordinates fibroblast proliferation and keratinocyte differentiation in wound healing via TGF-β-Slug signaling.

Vimentin has been shown to be involved in wound healing, but its functional contribution to this process is poorly understood. Here we describe a previously unrecognized function of vimentin in coordinating fibroblast proliferation and keratinocyte differentiation during wound healing. Loss of vimentin led to a severe deficiency in fibroblast growth, which in turn inhibited the activation of tw...

Activin B promotes BMSC-mediated cutaneous wound healing by regulating cell migration via the JNK-ERK signaling pathway.

Bone marrow-derived mesenchymal stem cells (BMSCs) are able to differentiate into various types of skin cells and participate in skin regeneration and repair. Activin signaling can regulate wound healing and reepithelialization. The present study assessed the impact of activin B on BMSC-mediated cutaneous wound healing in rats and explored the possible mechanism involved. We found that CFSE-lab...

12-hydroxyheptadecatrienoic acid promotes epidermal wound healing by accelerating keratinocyte migration via the BLT2 receptor

Leukotriene B4 (LTB4) receptor type 2 (BLT2) is a G protein-coupled receptor (GPCR) for 12(S)-hydroxyheptadeca-5Z,8E,10E-trienoic acid (12-HHT) and LTB4. Despite the well-defined proinflammatory roles of BLT1, the in vivo functions of BLT2 remain elusive. As mouse BLT2 is highly expressed in epidermal keratinocytes, we investigated the role of the 12-HHT/BLT2 axis in skin wound healing processe...

Ozone oil promotes wound healing by increasing the migration of fibroblasts via PI3K/Akt/mTOR signaling pathway

Skin injury affects millions of people via the uncontrolled inflammation and infection. Many cellular components including fibroblasts and signaling pathways such as transforming growth factor-β (TGF-β) were activated to facilitate the wound healing to repair injured tissues. C57BL/6 female mice were divided into control and ozone oil treated groups. Excisional wounds were made on the dorsal sk...
